摘要
为观察白介素 4单克隆抗体 (IL- 4Mab)对同种大鼠心脏移植物存活时间及局部细胞因子网络的影响 ,将实验对象分对照和实验组 ,分别静脉给予生理盐水及抗白介素 4单克隆抗体 (OX81) ,采用改良的移植术式建立移植模型 ,常规监测排斥反应发生情况。应用 RT- PCR的方法检测移植物局部细胞因子如 IL- 1β、IL- 2、CD2 5、IL- 4、IL- 5、IL- 6、IL-10、TNF- α、IFN- γ的表达水平。结果表明 :IL- 4Mab组移植心存活时间没有显著性变化 ,IL- 1β、IL- 2、IL- 5、CD2 5、TNF-α、IFN- γ的表达较强 ,而 IL- 4、IL- 6、IL- 10的表达较弱。提示单纯应用抗 IL- 4单克隆抗体可能调节 TH1/ TH2类 CK的表达 ,但对于移植物的存活时间却少有影响 ,其可能的机制为 :1CK网络中存在其它的独立于 IL- 4的 TH2类 CK;2TH亚群中存在除 TH1及 TH2亚群以外的亚群 THX;3IL-
To observe the influence of IL 4 monoclonal antibody (Mab) on allografts survival time and local cytokines network, the allografts were divided into two groups: control group receiving intravenous (i.v.) injection of normal saline (group A) and experimental group receiving injection (i.v.) of IL 4 Mab (group B) postoperatively. The local expression of cytokines IL 1β, IL 2, CD 25 , IL 4, IL 5, IL 6, IL 10, TNF α, IFN γ by using reverse transcriptsae polymerase chain reaction (RT PCR). The results showed in the group B, the survival time of the allografts was prolonged significantly, the expression of IL 1β, IL 2, CD 25 , IL 5, TNF α, IFN γ was up regulated, and the expression of IL 4, IL 6, IL 10 significantly down regulated as compared with group A. The results indicated that IL 4 Mab alone could regulate the expression of the Th type cytokines, but had a little influence on the survival time of the allografts.
