摘要 采用改变检测波长法 ,以 C18为固定相 ,以乙腈 -甲醇 - 0 .2 mol/L醋酸铵溶液 -水 ( 4 5∶ 1 0∶ 1 0∶ 35 ,p H=6.8)为流动相测定红霉素的含量。检测波长 :0~ 5 min(基线 )为 32 0 nm,5~ 8min(色谱峰 )为 2 1 5 nm。红霉素浓度在 0 .45~ 2 .7mg/ml范围内与峰面积呈良好的线性关系 ,回归方程为 Y× 1 0 -4 =0 .0 2 3+ 1 .2 38X,r =0 .9997,平均回收率 97.1 % An HPLC method was established for the assay of erythromycin. The stationary phase was Zorbax C 18 Column. The mobile phase consisted of methanal, acetonitrile, 0 2mol/L ammonium acetate solution and water(45∶10∶10∶35), adjusted pH value to 6 8 with acetic acid. The detector set at 320nm during first 5 minutes to get the gently base line and at 215nm during 5~8minutes to detect erythromycin level. The linearity was good in the range of 0 45~2 7mg/ml of erythromycin. The regression equation of erythromycin concentrations with the peak areas was Y×10 -4 =0 023+1 238X(r=0 9999). The average recovery was 97 1%.
