摘要
目的:在体外建立大鼠耳蜗螺旋神经节神经元培养模型,并提高其产出率及细胞活性。方法:将出生后5天的大鼠耳蜗螺旋神经节神经元在体外培养3~5天,用神经丝蛋白(Neurofilament protein,NFP)单克隆抗体进行免疫组化染色。结果:显示耳蜗螺旋神经节神经元在体外无血清培养条件下,可以存活并进行正常分化。结论:耳蜗螺旋神经节神经元在体外有稳定的可塑性及轴突再生修复能力。
To establish a cytological model of spiral ganglion neurons in vitro and to improve cell yield and viability. Methods: The spiral ganglia were dissected from five-day postnatal rats. The neurons were dissociated and cultured in serum-free medium for 3 - 5 days in vitro and then were immunostained with the monoclonal antibody against NEP. Results:The spiral ganglion neurons could survive and had a normal phenotypic differentiation in serum-free medium. Conclusion:The spiral ganglion neurons have stable plasticity and ability of neuritic regeneration.
