摘要
为探讨肝细胞生长素(HGF)促进肝细胞DNA合成的机理,结合Francavilla报告,血清雌二醇(E_2)水平能影响肝细胞DNA合成的研究,我们建立了SD大鼠急性肝损伤模型,并按不同的HGF剂量设实验组,不同时间取血检查血清E_2动态变化,同时用盐水设对照组。结果表明:大鼠肝损伤组注射HGF(20mg)后,E_2水平明显高于盐水对照组P<0.01;而血清E_2,浓度在4h达最高峰,并与剂量成正相关(γ_1=0.835,γ_2=0.885)。但E_2血清高峰期不因HGF剂量加大而提前。无肝损伤大鼠组:注射HGF和盐水对照组其血清均未检出E_2。提示HGF只能提高肝损伤大鼠血清E_2水平。推测HGF可能作用于肝损伤大鼠丘脑下部垂体而正反馈调节E_2水平,E_2再通过其相应受体效应调节钠通道,进而促进肝细胞再生。我们初步认为:肝损伤、HGF、E_2、肝细胞再生四者有密切关系。
In our laboratory, injection of HGF, prepared by the modified Labrecque's method, was used for the treatment of experimental hepatic injury in rats. It was found that the serum estradiol level was significantly higher in the group of HGF treatment than that in the control group (P<0.01), and the estradiol level, which increased in direct proportion to the dosage of HGF, reached the maximum 4 hours after the HGF injection in various dosages. Such a dynamic state did not appear in HGF treatment group without hepatic injury and in the NS control group. Thus we believe that HGF has a stimulative effect on the hypothalamus or the pituitary gland of the rats with liver injury, which regulates the serum estradiol level by positive feedback, and then hepatocyte regeneration is enhanced. It is our preliminary conclusion that there is a close correlation among liver injury, HGF, estradiol and the liver regeneration,
