家兔多器官功能衰竭发病过程中血浆血管活性肠肽的变化 被引量:2

STUDIES ON THE CHANGES IN PLASMA VIP CONTENT IN RABBITS WITH MOF

摘要 为探讨多器官功能衰竭(MOF)的发病机理,应用饲菌复合失血性休克再灌注损伤的方法复制家兔MOF模型,依诊断标准实验组17只动物有12只发生MOF(70.6%)。用放射免疫分析法检测了血浆血管活性肠肽(VIP)的动态变化。MOF动物(n=12)实验前血浆VIP含量为20.17±4.45ng/L,实验后1.5h明显升高(43.67±0.35ng/L),超过实验前1倍,相差显著(P<0.05),尔后逐渐下降,72h降至最低值(5.47±2.68ng/L),只有实验前的1/4,相差显著(P<0.05),同对照组(n=6)相差亦非常显著(P<0.01)。结果表明,VIP作为一种重要的胃肠调节肽,对MOF的发生发展具有明显的病理生理意义。 This report describes an animal model of multiple organ failure (MOF) in 17 rabbits. They were fed E. coli [5 × 1010/(ml·kg) B. W.], and then sustained severe hemorrhagic shock. 12 rabbits developed MOF from 1-3 days, a morbidity of 70.6%. Plasma vasoactive intestinal peptide (VIP) content in rabbits was assayed by means of radioimmunoassay (RIA). Before the experiment, the plasma VIP concentrations were 25.33±5.49ng/L (x±Sx) and 20.17 ±4.45ng/L in both control group (n = 6) and MOF group (n=12), respectively. No significant difference between the two groups (P>0.05) was found. The animals with MOF showed an obvious increase in plasma VIP at 1.5 hours (43.67± 10.35ng/L) after the experiment, which was significaltly higher than the original level (P<0.05). But the plasma VIP content returned to original level at 24 hours after the experiment. It still declined at 48 hours (15.72 ±4.26ng/L) and 72 hours (5.47±2.68ng/L) after the experiment. The value at 72 hours was only one fourth of the initial level, showing a conspicuous defferenee, and it was lower than that of control as well (P<0.01). This result suggested that VIP, as an important gastrointestinal peptide, played a role of adjustment function in the experimental MOF syndrome. The possible pathophy-siologic and clinical significance of these changes was discussed.
