去甲斑蝥酸钠干扰细胞有丝分裂的机制 被引量:15

摘要 本实验用去甲斑蝥酸钠10μg/ml处理CHO、HeLa和小儿包皮细胞6~24小时,观察到CHO和HeLa细胞胞质骨架(微管和微丝)结构有比较明显的变化。微管卷曲、缩短;微丝聚集成较粗的短纤维束,细胞变圆;M期纺锤体显著膨大,多极纺锤体比例上升。小儿包皮细胞的微管变化不明显,细胞外形变圆缓慢,24小时内可以自动恢复长梭形。纺锤体略微膨大,但仍然保持双极形式。去甲斑蝥酸钠通过引起细胞骨架异常,导致有丝分裂紊乱。其作用效应对非正常二倍体细胞(CHO)和肿瘤细胞(HeLa)大于对正常二倍体细胞(小儿包皮)。 CHO, HeLa and chinese fetal foreskin fibroblasts (F-cells)were treated with disodium norcantharidate (10μg/ml) for 6-24hr. In CHO and HeLa cells, the cytoskeleton (microtubules and microfilaments) changed obviously. The microtubles appeared more convolute in contour and then dispersed, the microfilaments gathered to form thicker filament bundles, and the cells turned round in shape, the spindles in M-phase expanded, more multipolar spindles appeared.On the other hand, the drug affected the cytoskeleton of F-cells slightly. The change of microtubules in contour was not obvious. The shape of F-cells turned round slowly, and returned longish spontaneously within 24hrs., the spindles of Mphase expanded more or less, but sill kept in bipolar position.By means of disrupting then cytoskeleton, disodium norcantharidate could result in disorder of mitosis and lead the CHO and HeLa cells to be killed.
