摘要
目的 探讨胎盘绒毛膜在胎儿期造血中的作用 ,寻找造血干 /祖细胞的新来源 ,供临床移植应用。方法 分别测定并比较脐动、静脉血中CD+34 细胞含量及粒 单细胞集落形成单位(CFU GM)、混合集落形成单位 (CFU MIX)、高增殖潜能集落形成单位 (HPP CFU)集落的产率。结果脐静脉中CD+34 及CD+34 /CD-38细胞含量分别为 (1 2 5± 0 94) %和 (0 6 1± 0 5 0 ) %,较脐动脉血中的含量(0 77± 0 5 0 ) %、(0 33± 0 2 7) %显著增高 (P均 <0 0 5 ) ;脐动、静脉血单个核细胞 (MNC)大多处于静止期 (>95 %) ,二者差异无显著性 (P >0 0 5 ) ,但合成期细胞的比率在脐静脉血中为 (2 2± 1 8) %,明显高于脐动脉血的比率 (0 9± 1 0 ) %;脐静脉血中CFU GM、CFU MIX和HPP CFU的产率分别为 (4 9 7± 2 8) / 2× 10 5MNC、(8 7± 4 6 ) / 1× 10 5MNC和 (1 6± 1 2 ) / 1× 10 5MNC ,明显高于脐动脉血的产率 (18±9) / 2× 10 5MNC、(5 2± 2 7) / 1× 10 5MNC和 (0 4± 0 4) / 1× 10 5MNC ,差异有显著性 (P均 <0 0 5 )。结论脐静脉血中造血干 /祖细胞的含量高于脐动脉 ,提示胎盘绒毛膜在胎儿期造血中具有一定的作用 ,并有可能成为造血干
Objective To study the role of chorionic villi of placenta on fetal hematopoiesis during embryo ontogeny, so as to find a possible new source of hematopoietic stem/progenitor cells (HSC/HPC) for clinical transplantation. Methods The umbilical cord blood was collected separately from the cord vein and the artery. The percentages of CD + 34 cells and it′s subtype were detected with fluorescence activated cell sorter (FACS) ( n =29), and the cell cycle distribution of MNC was also analyzed by FACS ( n =20). Granulocyte monocyte colony forming unit (CFU GM) ( n =11), mixed CFU (CFU MIX) ( n =8) and high proliferation potential CFU (HPP CFU) ( n =8) were respectively measured with semisolid agar culture and methylcellulose culture.Results The percentages of CD + 34 and CD + 34 /CD - 38 cells in cord vein blood were (1.56±1.91)% and (0 89±1 55)% respectively, which were significantly elevated as compared with those in cord artery blood (1 01±1 41)% and (0 60±1 40%), P <0 05. More than 95 percent of MNC in cord vein and artery blood was in stable stage. (2.15±1 84% of the cells was in synthesis stage in vein blood, which was higher than that in artery blood (0 9±1)%, P <0 05. The number of CFU GM, CFU MIX and HPP CFU were (50±28)/2×10 5MNC, (8.7±4 6)/1×10 5MNC and (1 6±1 2)/1×10 5MNC respectively, which were also higher in comparison with those in artery blood (18±9)/2×10 5MNC, (5.2±2.7)/1×10 5MNC and (0 4±0 4)/1×10 5MNC, P <0 05. Conclusion The number of HSC/HPC in cord vein was higher than that in artery. The results suggested that chorionic villi of placenta might play a role in fetal hematopoiesis, and it might be an alternative resource of HSC/HPC.
