摘要
目的 :建立人血清中异丙酚浓度的反相 HPL C测定法。方法 :以卡马西平为内标 ,采用 YWG- C1 8柱 ,甲醇 -水 (6 8∶ 32 )为流动相 ,检测波长为 2 5 8nm;流速为 1.0 m l/ min。结果 :异丙酚与内标分离良好 ,血清中异丙酚平均回收率为 10 1.2 % ,日内、日间 RSD均小于 10 % ,血清异丙酚浓度在 1~ 10︼g/ m l范围内线性关系良好 ,r=0 .9993。结论 :该法操作简便、结果可靠 。
AIM: To establish an assay method for propofol in human serum with reversed phase high performance liquid chromatography (RP HPLC). METHODS: Internal standard was carbamazepine, YWG C 18 was packed in the analytical column, The mobile phase consisted of methanol water (68∶32) and the flow rate was 1 0 ml/min. The UV detection wavelength was 258 nm. RESULTS: Propofol and carbamazepine were separated well. The mean recovery of propofol was 101 2%, and the ralative standard deviation (RSD) of within day and between day were all less than 10%, The assay linearity was determined over the range of 1-10 μg/ml in serum ( r = 0 999 3 ). CONCLUSION:The method is found to be sensitive with constant result. The experiments will supply reference methods for determination of propofol concentration.
