人预存天然抗体与猪胰腺细胞的异种反应性研究 被引量:2

Study on the xenoreactivity of human preformed natural antib odies with various porcine pancreatic cells

摘要 目的 分析人预存天然抗体与猪各种胰腺细胞的异种反应性及其同种型。方法 将患者和正常人血清预先加热灭活并按 1∶2、1∶4、1∶8、1∶16、1∶32倍比稀释 ,分别同猪胰腺冰冻切片于 37℃下孵育 30min ,冲洗后组织进一步用免疫荧光标记的羊抗人IgG和IgM抗体于 37℃下孵育 30min。采用间接免疫荧光染色法检测人预存天然抗体与猪胰腺细胞的反应性。结果  5 5 .6%的患者和 5 5 .0 %的正常人体内均存在着抗猪内分泌细胞的天然抗体 ,但患者血清中强阳性的百分率更高。 10 0 %的患者和 95 .0 %的对照血清都可同一种或多种胰腺细胞反应。预存天然抗体的同种型包括IgG和IgM两种。 结论 鉴于预存抗体同猪胰腺细胞的强阳性反应 ,移植前行组织配型试验和移植物预处理对移植的胰岛细胞的存活是必要的。 Objective To investigate the xenoreactiv ity of human preformed natural antibodies (PNAbs) with various porcine pancreati c cells and its isotypes.Methods Sera from healthy human and diabetic patients were heat inactivated and diluted by 1∶2, 1∶4, 1∶8, 1 ∶16, 1∶32, then incubated with pig cryostat sections for 30 min at 37℃. Aft er was hing, the tissues were further incubated with FITC-conjugated goat anti-human IgG and anti-human IgM antibodies for 30 min at 37℃. The reactivity of human preformed natural antibodies with various porcine pancreatic cells was determin ed by indirect immunofluorescence staining technique . Results 55.6% of IDDM patients and 55.0% of healthy human individuals contained preformed natural antibodies against porcine endocr ine cells. However, the percentage of strongly reacting sera in the patient grou p was significantly increased. All used sera from IDDM patients and 95.0% of ser a from healthy controls could react to one or the other of the various pancreatic cell types. The isotype of preformed natural antibodies showed both IgG and IgM .Conclusion In view of strongly positive reactivity of PNAbs with various porcine pancreatic cells, pretransplantly cross-matching test and graft pretreatment may be necessary for survival of transplanted islets.
