摘要
目的:评价体外5FU持续和间歇处理对Bel7402肝癌细胞株的生长抑制作用。方法:体外培养Bel7402肝癌细胞株,用一系列不同浓度的5FU对Bel7402肝癌细胞株分别进行每天24小时持续处理及2小时间歇处理,4天后进行MTT试验,检测肿瘤细胞的生长抑制率,对两组的IC50进行比较。结果:5FU处理浓度依次10.0μg/L、25.0μg/L、50.0μg/L、75.0μg/L、100.0μg/L、250.0μg/L、500.0μg/L、750.0μg/L、1000.0μg/L、2500.0μg/L、5000.0μg/L、7500.0μg/L、10000.0μg/L、20000.0μg/L持续处理组的肿瘤细胞抑制率依次分别为1.34%、2.01%、7.21%、10.37%、18.64%、29.96%、35.06%、43.35%、50.66%、87.63%、93.63%、94.46%、99.27%、99.35%,IC50是991.7μg/L。5FU处理浓度依次120.0μg/L、300.0μg/L、600.0μg/L、900.0μg/L、1200.0μg/L、3000.0μg/L、6000.0μg/L、9000.0μg/L、12000.0μg/L、18000.0μg/L、60000.0μg/L、90000.0μg/L、120000.0μg/L、240000.0μg/L2小时间歇处理组的肿瘤细胞抑制率依次分别为1.32%、1.78%、2.01%、2.89%、3.57%、5.94%、8.30%、10.36%、15.25%、17.27%、25.95%、32.63%、43.66%、67.81%,IC50为146600μg/L,为持续处理组的1478倍。
Objective: To evaluate the suppression effect of 5 Fluorouracil(5 FU) on Bel 7402 cell line by means of continuous or intermittent disposal. Methods: In vitro Bel 7402 cell line was cultured. A serials of different dosages of 5 FU were given to Bel 7402 cell line by means of 24 h continuous or intermittent disposal every day. 4 days later, MTT was taken to assay the inhibiting rates of the tumor cells. Results: The concentration of 5 FU was 10.0 μg/L,25.0 μg/L,50.0 μg/L, 75.0 μg/L, 100.0 μg/L, 250.0 μg/L,500.0 μg/L,750.0 μg/L,1 000.0 μg/L,2 500.0 μg/L,5 000.0 μg/L,75 000 μg/L,10 000.0 μg/L and 20 000.0 μg/L respectively. The inhibiting rates of tumor cells in continuous disposal group were 1.34%,2.01%,7.21%,10.37%,18.64%,29.96%,35.06%,43.35%,50.66%,87.63%,93.63%,94.46%,99.27%,99.35% respectively.The IC50 was 991.7 μg/L. The concentration of 5 FU was 120.0 μg/L、300.0 μg/L、 600.0 μg/L、 900.0 μg/L、 1 200.0 μg/L、 3 000.0 μg/L、 6 000.0 μg/L、 9 000.0 μg/L、 12 000.0 μg/L、18 000.0 μg/L、60 000.0 μg/L、90 000.0 μg/L、120 000.0 μg/L、240 000.0 μg/L respectively. The inhibiting rates tumor cells in 2 h intermittent disposal group were 1.32%,1.78%,2.01%,2.89%,3.57%,5.94%,8.30%,10.36%,15.25%,17.27%,25.95%,32.63%,43.66% and 67.81%,respectively. The IC50 was 146 600 μg/L. Conclusions: In vitro the inhibiting effect of 5 FU on Bel 7402 cells by means of continuous disposal with low concentration was higher than that of by means of intermittent disposal with high concentration.
