摘要
目的:观察γ射线诱导人内皮细胞凋亡及G-CSF对凋亡的保护作用。方法:对人脐静脉内皮细胞株ECV-304细胞γ射线照射后,以形态学方法和Comet分析观察凋亡的发生,并进行流式细胞仪定量分析。结果:γ射线能诱导内皮细胞凋亡,随时间延长凋亡增多,在0.5~15Gy范围内随剂量递增凋亡亦逐渐增加,而且照射后内皮细胞死亡以凋亡为主要形式。神经酰胺可模拟γ射线诱导的内皮细胞凋亡(32.11±1.33%),γ射线加TPA或G-CSF则抑制γ射线诱导的内皮细胞凋亡,分别为2.78±0.20%,2.77±0.25%,vs γ射线的13.80±0.46%(P<0.01)。结论:γ射线可诱导内皮细胞凋亡,并可被G-CSF抑制,其作用可能与细胞内神经酰胺和蛋白激酶C平衡的改变有关。
Objective:To investigate radiation-induced apoptosis of endothelial cell and its protection by G-CSF.Methods:The human umblical vein endothelial line cells(Ecs) were cultured and irradiated with γ-ray. The apoptosis was studied by morphological characteristics, Comet assay and quantitatively analysised with FCM. Results: Radiation(IR) induced apoptosis in Ecs which increased with postirradiation time and dose ranging from 0.5 Gy to 15 Gy. The apoptosis was the primary model of radiation - induced cell death of Ecs. Ceramide mimicked radiation to induce apoptosis(32.11±1.33%), while IR+TPA or IR+G-CSF inhibited radiation-induced appoptosis in Ecs, 2.78±0.20%, 2.77±0.35% vs IR 13.80±0.46%, respectively(P< 0.01). Conclusions: Radiation-induced apoptosis of Ecs may be inhibited by G-CSF and related to the variation of balance between ceramide and PKC signal systems.
