瘢痕疙瘩不同部位成纤维细胞的增殖-凋亡调控及相关蛋白的表达 被引量:1

摘要 目的 观察Fas介导下瘢痕疙瘩不同部位成纤维细胞增殖与凋亡的特点,探讨瘢痕疙瘩周边和中央部生长差异的细胞生物学机制。方法 应用透射电镜及流式细胞仪技术,观察了FasMcAb诱导下瘢痕疙瘩不同部位成纤维细胞的凋亡情况,并检测了不同来源成纤维细胞Fas受体、Bcl-2蛋白及p53蛋白的表达情况。结果 (1) 瘢痕疙瘩不同部位成纤维细胞在各种浓度梯度的FasMcAb作用下均未见明显的凋亡现象,Fas受体均呈高表达;(2)Bcl-2蛋白在瘢痕疙瘩不同部位成纤维细胞内均呈低表达,但中央部表达强度显著高于周边部(P<0.05);(3) p53蛋白在瘢痕疙瘩周边呈低表达,而在中央部呈高表达。结论 p53蛋白可能是导致瘢痕疙瘩不同部位呈现不同生长特性的最重要的增殖调控蛋白。 : Objective To explore the machanism which accounts for the different growth behavior observed in the peripheral and central areas of the keloids. Methods The fibroblasts derived from keloids were exposed to anti-Fas mAb at different concentrations to investigate Fas-induced apoptosis by means of transmission electron microscope and flow cytometry. The expression of Fas、Bcl-2 and p53 proteins was measured. Result No obvious apoptosis in response to anti-Fas mAB treatment was detected in the fibroblasts, while high expression of Fas protein was observed. Low Bcl-2 protein expressions were universal in the fibroblasts from the keloids, but thosed from the central area of the keloids manifested higher expression compared with the periphal fibroblasts (P<0.05). The expression of p53 protein was significantly elevated in the central fibroblasts in comparison with the peripheral ones. Conclusion p53 protein may account for the varied growth behavior as seen in different areas of the keloids.
