摘要
目的 观察参附注射液对吗啡依赖人神经母细胞瘤细胞SK-N-SH胞内Ca2+浓度的影响,探讨其戒毒作用的细胞机制。方法 建立吗啡依赖神经细胞模型,Fluo-3着染细胞后,应用流式细胞仪测定参附注射液对神经细胞内Ca2+浓度的影响。结果 参附注射液中、高剂量组神经细胞内Ca2+浓度显著升高,纳洛酮催促引起的细胞内Ca2+的降低抑制。 结论 参附注射液可能通过调节细胞内Ca2+浓度发挥其戒毒作用。
: Objective To observe the effect of Shenfu injection on intracellular calcium concentration in morphine-dependent human neuroblastoma SK-N-SH cell line, and explore its cellular mechanism of controlling drug-dependence. Methods Morphine-dependent cell line was estabilished by administering morphine at different doses into the culture medium. The cells were stained with fluo-3, and the intracellular calcium level was measured by using flow cytometry. Result Middle and high dosages of Shenfu injection could elevate the intracellular calcium level and inhibit the decrease of Ca2+ induced by naloxone. Conclusions The mechanism of counteracting drug-dependence of morphine with Shenfu injection might involve the regulation of intracellular calcium concentration.
