摘要 目的 :探讨实验性铅中毒对小鼠附睾神经生长因子 (NGF)基因表达的影响。方法 :小鼠以醋酸铅染毒 ,采用原子吸收分光光度法测定其血铅及附睾组织铅含量 ;通过免疫组织化学法和RT PCR法 ,观察附睾组织NGF和NGFmRNA含量的变化。结果 :铅染毒小鼠血铅和附睾组织铅含量显著升高 ;免疫组化结果显示 ,染毒组小鼠附睾管上皮NGF免疫反应明显减弱 ;RT PCR结果表明 ,铅染毒可使附睾组织NGFmRNA含量显著下降 ,而且随着染毒时间的延长 ,NGFmRNA含量呈指数下降。结论 :铅可使附睾NGF基因表达下降。 To study the effect of lead exposure on expression of NGF gene in mouse epididymis. Methods: Mice were exposed to acetate lead. The lead contents of blood and epididymis were determined by atomic absorption spectrometry. The contents of epididymis NGF and NGF mRNA were analyzed by immunohistochemistry and RT-PCR method. Results: The lead contents in blood and epididymis of lead exposure were distinctly increased. The reaction of immunhistochemistry indicated that NGF-like immunoreactivities in epididymis epithelium of lead exposure were reduced significantly. It was demonstrated that the expression levels of NGF mRNA in epididymis of lead exposure by RT-PCR method were decreased. Following the increase of lead exposure time, the expression of NGF mRNA in epididymis had a tendency to decrease in an index. Conclusion: Lead can reduce the expression of NGF gene in epididymis.
