摘要
将兔 2 细胞 ,4 细胞 ,8 细胞以及桑椹期的胚胎快速冷冻。以乙二醇作低温保护剂 ,浓度分别为 0 5 ,1 5 ,2 5 ,3 5mol/L。其中含 1 0 %犊牛血清和 0 2 5mol/L蔗糖 ,培养到囊胚阶段。结果发现 ,当乙二醇的浓度大于或者等于 2 5mol/L时 ,保护效果较好。在囊胚之前 ,发育程度越高 ,抗低温的能力越强 ,胚胎的发育率也就越高。
The rabbit embryos at stages of 2 cell,4 cell,8 cell and morula were frozen with vitrified methods.Ethylene glycol was used as cryoprotectants in concentration of 0 5,1 5,2 5 and 3 5 mol/L,respectively,and with 10% calf serum and 0 25 mol/L sucrose each.The embryos were cultured in vitro.The rates of development to the stage of blastocyst were improved with the increasing of cryoprotectant concentrations from 0 5 to 3 5 mol/L.The abilities of enduring low temperature were enhanced with embryo development before blastocyst stage.The blastocysts treated in different stages developed into normal individuals after embryo transfer.
