电穿孔法转化大肠杆菌TG1的条件优化 被引量:7

摘要 研究了质粒pFab74X经电穿孔转化大肠杆菌TG1菌株的影响因素 ,并优化其转化条件。电场强度和脉冲时间的影响较为复杂 ,两者适当组合才可获得高转化效率。此外 ,采用对数生长早期的细菌 。 Electroporation was performed to transfer plas mid DNA of pFab74X into Ecoli strain TG 1.A number of factors that effected transformation effi ci ency were investigated.High transformation efficiency was obtained with an app ropriat e combination of field strength and pulse time.Transformation efficiency could also be elevated by using hosted cells harvested in the early exponential growth phase, increasing the density of cells or decreasing the concentration of Ampci llin in the course of screening
