摘要
建立了适合中国国情的大规模牛血红蛋白 (Hb)的提取和纯化方法 ,为血液替代品的研究和生产提供原料保障。通过建立水性两相系统提取法、乙酸锌沉淀提取法、CO饱和提取法和板框过滤结合脱氧加热提取法 ,比较了用不同方法提取的牛Hb的纯度和牛Hb中所含的metHb、HbCO和HbO2 的含量。板框过滤结合脱氧加热提取法对器材要求不高 ,便于产业化 。
A suitable method of purifying large scale bovine H b,that can supply enough raw material for the research and manufacture of blood substitute was studied.Different bovine Hb purification me thods,such a s aqueous two-phase system,zinc acetic precipitation,carbon monoxide saturati o n and filtrative fiber-board filtration combining deoxygeningly heating pu rifica tion method were tested.Results showed that the fiber-board filtration combining deox y geningly heating method could prepare large scale bovine Hb with high purityIn addition the equipment could be easily prepared,so it is a suitable route to make raw material for the research and manufacture of blood substitute
