摘要
目的 建立反相高效液相色谱测定天麻钩藤制剂中钩藤碱和黄芩苷的方法。方法 采用ShimpackCLCODS柱 ,流动相为乙腈 水 草酸 SDS[4 5 0mL∶5 35mL∶5mL(0 .5→ 5 )∶10mL(2→ 10 ) ]和磷酸 (1→ 146 ) 乙腈 (18∶7) ,检测波长 2 5 0和 2 77nm ,进样量 10 μL。结果 线性范围 0 .2~ 1.0 μg和 0~ 1.0 μg ,相关系数为 0 .9991和 0 .9998,平均回收率钩藤碱 96 .82 % ,黄芩苷97.42 % ,日内RSD分别为 :0 .49%和 0 .2 6 %。结论 方法简便 。
OBJECTIVE: To establish a method for the determination of two contituents in Tianma Gouteng Yin by reversed-phase high performance liquid chromatography. METHODS: The chromatographic conditions included shimpack CLC ODS column and the mobile phase consisting of a mixture of acetonitrile-water-oxalie acid-SDS[450 mL:535 mL:5 mL(0.5&rarr5):10 mL(2&rarr10)] for rhynchophylline and phesphoric acid(1&rarr146)-acetonitrile (18:7) for baicalin. The detection wavelengths were at 250 nm and 277 nm, respectively. RESULTS: A good linearity was found over the concentration range of 0.2 ∼ 1.0 μg for rhynchophylline and 0 ∼ 1.0 μg for Baicalin, respectively. The correlation coefficients were 0. 9991 and 0. 9998, respectively. The average recovery of rhynchophylline was 96.82%, and that of baicalin was 97.42 %. The within-day RSD was 0.49% and 0.26%, respectively. CONCLUSION: The method was found to be simple and accurate.
