摘要
本文介绍一种新型光学蛋白质芯片技术和在生物医学领域的应用。通过固体基底表面的微格式化、表面改性和生物配基固定等技术 ,制备多元生物活性感应表面 ;利用生物分子操作、蛋白质的特异结合性和高分辨率的生物光学椭偏显微成像技术 ,达到识别、检测和纯化蛋白质的目的。它是一种非标记的多元蛋白质定量分析方法 ,并且能够对蛋白质之间的相互作用过程进行实时测量 。
One kind of optical ProteinChip, and a ProteinChip analysis system, as well as some applications are introduced in this paper. The chip is prepared with surface patterning, surface modification, ligand immobilization to form a sensing surface with multi-bioactivity. The affinity between proteins, and the optical imaging ellipsometry with high spatial resolution are used to realize non-labeling micro-assays for protein identifications and protein interactions. It is not only a qualitative, but also a quantitative way for protein interaction determination. It is able to obtain dynamic data of protein interaction process, such as interaction rates and conditions, with real-time detection.
