BACTEC MGIT 960全自动分枝杆菌培养鉴定药敏仪的动物实验模型分析

摘要 利用BACTEC 96 0全自动分枝杆菌培养鉴定药敏仪建立动物试验模型 ,以观察结核分枝杆菌 (M .tuber culosis.MT)MPT6 4质粒DNA疫苗预防结核病的效力。第一组小鼠接受H37RV攻击一个月后的肺、肝、脾培养结果 :90 %阳性标本培养时间为 7~ 13天。肝脏仅 75 %出现阳性结果。第二组小鼠接受H37RV攻击两个月后 ,其肺、肝、脾培养结果 :90 %阳性标本培养时间为 10~ 15天 ,肝脏仅 6 0 %出现阳性结果。第二组比第一组生长天数总体向后推迟 ,生长时间明显集中 ,生长曲峰强度比第一组明显减弱 ,第二组在肝脏中的感染率比第一组有所下降。可能是因为小鼠经DNA疫苗刺激后 ,免疫力增强 。 To observe the preventative effects to MPT64 DNA vaccine Mycobacterium tuberculosis, the experimental animal models were estabelished by using BACTEC-960 culture system. 16 BALB/C mice in the first group and 20 BALB/C mice in second group were challenged with H 37 Rv. The culture results in lung, liver and spleen were showed for the first group after one month and for the second group after two monthes. The results of 90% positive samples showed that the positive rate in liver 7~13 days for the first group was 75% and in 10~15 days for the second group was 60%. Compared with the first group, the second group had a long and concentrative growth time, at the same time, the peak intensity of growth cure was weakened and the infective rate in liver was falled off. The reason may be that the immunological enhancement of mice challenged with DNA vaccine slowed down the growth of Mycobacterium tuberculosis.
