摘要
目的 :研究亚低温复合黄芩素甙对脑缺血后沙土鼠海马组织能量代谢和病理学的影响。方法 :采用沙土鼠前脑缺血模型 ,缺血时间 10分钟。于再灌注 6 0分钟测定 ATP及腺苷酸池的含量 ,再灌注第 4日进行组织病理学检查。结果 :常温组 ATP含量〔(0 .5 1± 0 .19) mm ol/g〕较假手术组明显降低〔(0 .97± 0 .11) m mol/g,P<0 .0 1〕,亚低温组〔(0 .73± 0 .0 8) mm ol/g〕或黄芩素甙组〔(0 .70± 0 .0 8) m mol/g〕均高于常温组 (P均 <0 .0 5 ) ,亚低温复合黄芩素甙组 ATP含量〔(0 .83± 0 .0 5 ) mmol/g〕明显高于亚低温组或黄芩素甙组 (P<0 .0 5和 P<0 .0 1) ;海马 CA 1区存活神经元计数亚低温组或黄芩素甙组均较常温组明显增多 (P均 <0 .0 1) ,而与亚低温复合黄芩素甙组相比则明显减少 (P均 <0 .0 1)。结论 :亚低温复合黄芩素甙能减少脑缺血后海马 CA1区锥体细胞延迟性死亡 。
Objective:To investigate the effects of hypothermia combined with breviscarpin on energy metabolism following transient forebrain ischemia in gerbils.Methods:The ischemia was induced by occlusion the bilateral common cartoid arteries for 10 minutes.Histopathology test was carried out at the fourth day.ATP contents were detected at 60 minutes after reperfusion.Results:The ATP contents in normothermia(NT) group 〔(0.51±0.19)mmol/g〕 were lower than that in sham (Sham) group 〔(0.97±0.11)mmol/g〕,and the ATP contents in hypothermia(HT) group 〔(0.73±0.08)mmol/g〕 and breviscarpin (Bre) group 〔0.70±0.08)mmol/g〕 were higher than that in NT group(both P<0.05),and the ATP contents in hypothermia combined with breviscarpin(HB) group were higher than that in HT or Bre group(P<0.05 and P<0.01).Animals of HT or Bre group showed less neuron loss of CA1 than that of NT group(both P<0.01),but more neuron loss than that of HB group(both P<0.01).Conclusions:Hypothermia combined with breviscarpin can decrease delayed neuronal death in hippocampal CA1 region,its mechanism may be reducing ATP depletion.
