摘要
目的 :研究黄芪注射液对缺血再灌注 (IR)性肾损伤肾组织中白介素 6 (IL 6 )和碱性成纤维细胞生长因子 (b FGF)的影响 ,探讨黄芪注射液减轻 IR肾损伤的机制。方法 :成年日本大耳白兔 2 4只 ,随机均分为手术对照组、单纯缺血再灌注组 (IR组 )和黄芪注射液处理组 (黄芪 +IR组 )。用右肾动脉夹闭法制备肾 IR损伤模型。观察肾组织超微结构的改变 ,检测血清肌酐 (SCr)含量和肾组织中 IL 6、b FGF含量。结果 :肾缺血 1小时再灌注 48小时 ,电镜下观察 IR组肾组织变性改变显著 ,黄芪 +IR组病变明显减轻 ;SCr含量和肾组织中IL 6含量 IR组高于手术对照组 ,黄芪 +IR组低于 IR组 ,差异均有显著性 (P<0 .0 5或 P<0 .0 1) ;肾组织中b FGF含量 IR组低于手术对照组 ,黄芪 +IR组高于 IR组 ,差异均有显著性 (P<0 .0 5或 P<0 .0 1)。结论 :IR肾损伤时肾组织中 IL 6含量升高、b FGF含量降低 ;黄芪可能通过调控 IL 6和 b FGF的产生或破坏而减轻 IR肾损伤。
Objective:To investigate the effects of Astragalus membranaceus injection (黄芪注射液) on content of interleukin6(IL6) and basic fibroblast growth factor (bFGF) in rabbit kidneys with ischemia reperfusion (IR) and to explore the mechanism of Astragalus membranaceus injection in protecting kidney from IR injury.Methods:Twentyfour adult Japanese white rabbits were randomly divided into operational control group,IR group and Astragalus membranaceus injection treatment group (AM+IR group) in the study.The rabbit's left kidney was removed,and artery of right kidney was clasped for one hour and then reperfused 48 hours.The content of serum creatinine (SCr) and content of IL6 and bFGF in the kidney were measured.And the changes in renal structure were detected under electron microscope.Results:The degenerative changes were remarkable under electron microscope in renal tissue in IR group,but the changes in AM+IR group were slightly.The content of SCr and IL6 in kidney in IR group was much more than those in operational control group,and lower than those in AM+IR group,there was a significant difference between these groups (P<0.05 or P<0.01).The content of bFGF in kidney in IR group was much lower than those in operational control group and AM+IR group(P<0.05 or P<0.01).Conclusions:The content of IL6 in kidney was increased and the content of bFGF decreased after kidney ischemia reperfusion;AM has the effect of alleviation the renal IR injury by modulation the production of IL6 and bFGF.
