摘要
[目的]研究钻的生殖毒性。[方法]采用卵母细胞体外培养和体外受精的方法。[结果]氯化钴降低体内(0h)第一极体释放率和卵母细胞存活率(P<0.01),降低卵母细胞(24h)的体外受精率(P<0.01),且有剂量依赖性增加,随着体外培养时间的延长,2个剂量组的第一极体释放率与各自0h相比有显著提高,除了6.0mg·kg^(-1)组外,48h后3.0mg·kg^(-1)剂量组的IVF率与24h时相比显著提高,3.0mg·kg^(-1)和6.0mg·kg^(-1)两组的IVF率在48h和72h时与对照组比差异均有显著性(P<0.05,P<0.01)。[结论]实验结果提示,氯化钴能够抑制卵母细胞的体内成熟,降低卵母细胞的体外受精能力,具有明显的生殖毒性。
[Objective] To study the reproductive toxicity of cobalt. [Methods] Adopt the method of vitro culture and vitro fertilization (IVF) of mouse oocyte. [Results]Cobalt chloride can inhibit the extruding of the first polar body and affect the quality and the viability of mouse oocyte and reduce the rate of IVF, while has little impact on germinal vesicle breakdown(GVBD)of mouse oocytes in vivo. By the culture of mouse oocytes and zygotes in normal culture solution (DMEM + BSA, CZB) the extruding of the first polar body compared with o h respectively and IVF rates in 3 .0 groups increased compared with 24 h respectively. But they also appeared to have significant difference compared with contrast group. [Conclusion]The result indicates that cobalt chloride may be hazardous to meiosis maturation of mouse oocyte which reduced the ability of fertilization and may be toxic to women oocyte and reproductive capacity.
