摘要
以体外培养的大鼠胰岛细胞为对象 ,利用放射免疫检测技术、免疫组织化学技术及透射电镜观察低剂量PrCl3对胰岛 β细胞分泌胰岛素功能的影响。当PrCl3浓度分别为 0 0 1和 0 1mmol·L- 1 时 ,培养液中胰岛素含量及培养孔内β细胞数目明显高于对照组 。
The effects of low dosage PrCl 3 on the rat islet cell in vitro and the mechanism for the insulin release were investigated. Cell culture of rat pancreas islets in media with or without low dosage PrCl 3 was studied. The concentration of insulin was determined by radioimmunoassay. The cultured cells were observed by immunohistochemistry and transmission electron microscopy. The results indicate that the levels of insulin in the experiment groups are higher than that in the control groups. Insulin and the number of β cell increase significantly at 0 1 and 0 01 mmol·L -1 PrCl 3 groups. There are also some stem cells around the β cells in the 0 1 and 0 01 mmol·L -1 PrCl 3 groups. PrCl 3 improves β cell to secrete insulin directly at low dosage and may stimulate the stem cells to differentiate into β cells.
