摘要
目的 :以雷洛昔酚 (Raloxifene)和维生素 D3(VD3)为对照 ,研究补肾益精方在治疗骨质疏松症 (Os-teoporosis)中对肌球蛋白重链基因 (MHC- ) m RNA的表达和骨密度 (BMD)的影响。方法 :40只 1 0月龄Wistar雌性大鼠分五组 :正常组 (卵巢假切除 )、卵巢切除组、补肾益精方组 (卵巢切除 +补肾益精方 )、Ralox-ifene组 (卵巢切除 +Raloxifene)、VD3组 (卵巢切除 +VD3)。卵巢切除术后 90天给药 ,给药 90天后处死。取股四头肌、股骨、血清分别运用 RT-PCR法、DPX-L型骨密度仪、放射免疫法测定 MHC- 、骨密度、雌二醇(E2 )水平。结果 :(1 )正常组 (假手术组 ) MHC- 电泳条带清晰可见 ,而卵巢切除组的 MHC- 表达未达到本实验方法检测水平 ,虽然经多次重复均不显现电泳条带。(2 )与卵巢切除组相比较 ,补肾益精方、Ral和 VD3组均显著提高了四头肌 MHC- m RNA表达水平 ,电泳图谱均呈现清晰的条带 ,显示了药物的逆转作用。3个用药组的上调作用无统计差异 (P>0 .0 5 )。 (3 )与卵巢切除组相比较 ,3个用药组的股骨 BMD均显著增加 (P<0 .0 5 ) ,也显示了药物的逆转作用 ,但相互间也无统计差异 (P>0 .0 5 )。结论 :补肾益精方、Raloxifene和 VD3都能对卵巢切除大鼠股四头肌中 MHC- 的表达和股骨 BMD具上调作用。
Objective:effect of BuShenYiJingFang on bone mineral density(BMD) of femora and the expression of typeⅠmyosin heavy chain(MHC-Ⅰ) gene mRNA of quadriceps femoris were investigated in ovariectomized rats in comparison with that of Raloxifene (Ral) or Vitamin D_3 (VD_3). Methods : 90 days after ovariectomy, thirty-two 10-month-old female Wistar rats were randomized into 4 groups(8 rats/group):Ovariectomy group(OVX);BuShenYiJingFang group that were treated with 0.39g of BuShenYiJingFang /kg/d;Ral group and VD3 group that were treated with 4.54 mg of Ral/kg/d and 0.04 mg of VD_3/kg/d respectively.The other eight rats were sham-operated.90 days after administration, all rats were sacrificed and MHC-Ⅰgene mRNA of quadriceps femoris ?BMD of femora and oestradiol(E_2) in serum were determined respectively with a reverse transcriptase-polymerase chain reaction assay(RT-PCR)?instrument of measuring BMD and radio-immunity assay.Results: (1) The MHC-Ⅰgene mRNA electrophoresis bands of sham-operated group rats were clear. But the bands of OVX group did not emerge these bands by repeated test. Its expression hadn't reach detective level. (2) BuShenYiJingFang ?Raloxifene and VD_3 all significantly increased the expression of MHC-Ⅰgene mRNA of quadriceps femoris for ovariectomized rats (P<0.05) contrast to that of OVX group. Their bands were clear too. The up-regulated effect of three drug groups hadn't statistical diference(P>0.05). (3) The femurs BMD of three drug group rats were significantly increased (P<0.05) contrast to that of OVX group. But the BMD among three drug groups hadn't statistical diference (P>0.05). Conclusion: These results showed that BuShenYiJingFang?Raloxifene and VD_3 can increase the expression of MHC-Ⅰof quadriceps femoris and BMD of femora in ovariectomized rats. By statistical comparison, there were no significant differences for MHC-Ⅰgene expression and femurs BMD among all drug groups and for E_2 level between BuShenYiJingFang and Ral groups. So three drugs can reversed respectively the effect of ovariectomy for the expression of MHC_Ⅰand BMD.
