摘要
目的 :骨碎补是骨伤科常用的一味中药 ,为了进一步探讨其促进骨折愈合的机理 ,设计了该实验以期为临床运用骨碎补提供理论依据。方法 :将 3 6只 SD大鼠随机分成三组 :生理盐水组 (A)、骨碎补组 (B)、伤科接骨片组 (C)。按标准骨折模型方法造模 ,不作任何外固定 ,分笼饲养。从手术后第二天开始灌胃给药 ,骨碎补组每天以骨碎补水煎液分早晚 2次灌胃 ,伤科接骨片组每天给药 2次 ,对照组同时给予等量生理盐水。于术后第 7天、1 4天、2 1天每组分别处死动物 4只 ,取出骨标本。骨标本制成切片测量骨痂厚度及 TGF-β1 免疫组化染色。结果 :骨碎补组骨痂组织形成丰富 ,厚度明显优于对照组 (P<0 .0 1 ) ;骨碎补组骨痂组织中 TGF-β1表达量及阳性定位与伤科接骨片组相同 ,均明显强于对照组。结论 :实验结果表明骨碎补能增加骨痂厚度 ,提高骨折愈合质量。增加 TGF-β1 在骨痂组织中的表达 ,可能是骨碎补促进骨折愈合的机制之一。
Objective:This experiment has been designed to study the mechanism of Gusuibu's promoting the healing of fracture and provide the better theoretical basis for more extensive clinical application.Methods:Thirty-six SD rats were divided into three groups at random and equally:Group A(control),Group B(Gusuibu),Group c(shangke jiegupian). Standard fractures were made at middle sments of right tibias in thirty-six rats. At the first, second, third weeks after fracture, bone specimen was collected from every rat. Then measuring thickness of calus and immunohistochemical studies were performed on every slice.Results:During fracture healing, the thickness of callus and expressions of TGF-β_1 in callus in Group B were much better than in control.Conclusion:According to the results, we maintain the mechanism of Gusuibu on promoting fracture healing:enhance the expression of TGF-β_1 in callus.
