摘要
目的 探讨恶性疟原虫环子孢子蛋白基因在耻垢分枝杆菌M Smegmatismc2 15 5中的表达。方法 采用电穿孔转化法将重组大肠杆菌 -分枝杆菌穿梭表达质粒 pBCG/CSP导入耻垢分枝杆菌M Smegmatismc2 15 5 ,重组分枝杆菌培养4 8- 72h后于 4 5℃诱导表达 3 0min ,表达产物进行SDS -PAGE及免疫印迹分析。结果 SDS -PAGE及免疫印迹分析结果均显示在约 4 2kDa的位置上可见明显的蛋白条带。结论 恶性疟原虫环子孢子蛋白可在分枝杆菌中表达。
Aim To explore the expression of the circumsporozoite protein(CSP) gene of Plasmodium falciparum FCC-1/HN isolate in Mycobacteria Smegmatis mc 2 155 Methods The recombinant E coli-Mycobacteria shuttle plasmid pBCG/CSP was introduced into M Smegmatis mc 2 155 by electroporation,the recombinat Mycobacteria was induced at 45 ℃ for 30 min after 48-72 h culture,and its expression product was analysed by SDS-PAGE and Western-blot Results The expressed CSP could be observed in by SDS-PAGE and Western-blot with the molecular weight of 42 kDa Conclusion The CSP gene could be expressed in Mycobacteria Smegmatis mc 2 155
