摘要
研究了新型镇痛药盐酸曲马多的重要中间体 2 -二甲胺甲基环己酮的合成。通过正交实验 ,找到了最佳反应条件。最佳反应条件为 :环己酮、聚甲醛、二甲胺盐酸盐配料摩尔比为 2 :1 :1 ,反应温度 95℃ ,反应时间 2 .5 h,收率为 74.1 % .并通过红外光谱。
The synthesis of 2 (dimethylamino)methyl cyclohexanone which is Mannich base has been studied. It is an important intermediate of the total synthesizing the new analgesic tramadol HCl. The optimum reaction conditions have been found by orthogonal test .The result show that the yield can reach 74.1% under the optimum conditions,which means cyclohexanone:polyformaldehyde:dimethylamine hydrochloride=2∶1∶1(molar ratio), reaction temperature is 95℃,reaction time is 2.5 hours.The product is analyzed qualitatively by means of IR and 1HNMR.
