环腺苷酸应答元件结合蛋白(CREB)参与人hsp90β基因的热诱导表达 被引量：2

摘要 为研究ｃＡＭＰ应答元件（ＣＲＥ）在人热休克蛋白ｈｓｐ９０β基因表达中的作用，构建了数个受ｈｓｐ９０β基因不同长度调控片段介导的氯霉素乙酰基转移酶（ＣＡＴ）报告基因质粒．分别转染Ｊｕｒｋａｔ细胞并检测４２℃１ｈ热诱导前后 ＣＡＴ活性．结果发现删除ｈｓｐ９０β基因上游含ＣＲＥ片段（－１７３／－９１ｂｐ）后，ＣＡＴ的热诱导表达活性降低了６７％．电泳迁移率变更分析（ＥＭＳＡ）显示热休克后Ｊｕｒｋａｔ细胞的核抽提物中磷酸化的ＣＲＥＢ与该片段呈特异性结合．Ｗｅｓｔｅｒｎ印迹实验及ＰＫＡ活性检测发现ＣＲＥＢ的磷酸化程度及ＰＫＡ活性均随热诱导时间的延长而增加．以上结果表明：磷酸化的ＣＲＥＢ通过与ｈｓｐ９０β因上游的ＣＲＥ结合参与该基因的热诱导表达，并可能与ＰＫＡ传导途径有关．
