TPA 对人早幼粒白血病细胞系 HL-60诱导分化的超微结构研究 被引量:1

摘要 对经 TPA 诱导分化的人早幼粒白血病细胞系 HL-60细胞在透射和扫描电镜下进行了观察。TPA 诱导后 HL-60细胞向巨噬样细胞分化,在诱导后1d 大部分细胞已在细胞形状表面构型和突起等方面出现了变化。细胞核、细胞膜及线粒体等细胞器均有改变。TPA 的亲脂性与细胞膜相结合有可能改变细胞受体的构型。推测线粒体等的明显变化将导致细胞的能量代谢、蛋白质合成活动的障碍,而且可能通过抑制肿瘤细胞的能量代谢和蛋白质合成系统促使白血病细胞发生退变和死亡。 The induction of differentiation of human promyelocytic cell line HL-60 cells by 12- O-tetradecanoyl-phorbol 13-acetate (TPA) is reported. Several ultrastructural changes were observed under transmission electron microscope (TEM) and scanning electron microscope (SEM). When HL-60 cells were incubated with TPA for 1-2 days,most of them differenti- ated into macrophage-like cells.After 24 h exposure to TPA,HL-60 cells showed a wide variety of shapes with long thin processes or squirrel tail-like processes extending from the surface of the cell.Some morphological changes in cell surface configuration were also ob- served.After 48 h treatment with TPA some changes of organelleswere found.The elec- tron density of mitochondrial matrix was increaed.The cristae of mitochondrion condensed and some of them showed feature of myelin figures. Besides,the invaginations of nuclear membrane,lysosomes and osmiophilic masses were increased.The rough endoplasmic reticulum showed fragmentation,and the free ribosomes decreased in number. It is possible that the lipophilic TPA by itself after being bound to the cell membrane, can also modify the configuration of other receptors. We consider that the prominent morphological changes of mitochondria,rough endo- plasmic reticulum might lead to the derangement of cell energy metabolism and synthetic activity causing degeneration and death of leukemic cells.
