摘要
本文报道从选择细胞、摸索出斑条件开始,采用 H_(8205)株、118/76株及美国草原田鼠体内分离的 PH 病毒和流行性肾病(NE)等肾综合征出血热(HFRS)相关的病毒株,在 Vero E_6细胞上建立界限清楚呈圆形的空斑,空斑直径0.2~3mm,6~10d 出齐,空斑数与接种病毒剂量成正比,两者几乎呈直线相关。空斑可被高价特异性血清所中和,已建立的空斑法适于 HFRS 病毒的定量工作。
The 118/76,H_(8205) strains of Haemorrhagic fever with renal syndrome (HFRS) and the nephropathy epidemic (NE) and Prospect Hill (PH) viruses were detected on mo- nolayers of Vero E_6 cells by plaque method using agarose overlay medium.The sizes of circular plaques formed 6 to 8 days post-infection were 0.2-3mm in diameter.Plaque counts were reproducible and an approximately linear relationship between the number of plaques and the virus concentration of the inocula existed.A neutralization reaction of the plaques showed its specificity.
