心房利钠多肽的规模化制备及活性分析

Large Scale Preparation and Activity of Atrial Natriuretic Polypeptide

目的 建立一种简便、有效的心房利钠多肽（ａｔｒｉａｌ ｎａｔｒｉｕｒｅｔｉｃｐｏｌｙｐｅｐｔｉｄｅ，ＡＮＰ）的规模化制备方法。 方法 以恒河猴心房为原料，采用匀浆、分级超滤制备ｍ－ＡＮＰ，用ＳＤＳ－ＰＡＧＥ测定纯度和相对分子质量，用Ｗｉｓｔａｒ大 鼠检测生物学活性。结果 所制备的ｍ－ＡＮＰ相对分子质量主要分布在２０００～６０００，包含了ＡＮＰ的主要活性成分。 具有降低血压，扩展血管和利钠利尿的生物学活性。结论 该工艺可用于规模化生产ＡＮＰ。

Objective To establish a simple and effective method for the large scale preparation of at-rial natriuretic polypeptide( ANP) from animal hearts. Methods m-ANP was prepared form the hearts of rhesus by fractional ultrafiltration, and the purity and relative molecular weight of it were detected by SDS-PAGE . The biological activity of it was ananlyzed with Wistar rats. Results The relative molecular weitght of main active components of the m-ANP ranged from 2,000 to 6,000. The m-ANP showed the biological activities of lowing blood pressure, dilating blood vessels, natriuresis and diuresis. Conclusion The method is suitable for the large scale preparation of ANP from animal heats.
