河南汉族人群9个STR基因座的遗传多态性

摘要 目的 研究河南地区汉族CSF1PO、TPOX、TH0 1、F13A0 1、FESFPS、vWA、D16S5 39、D7S82 0、D13S317等 9个短串联重复序列 (STR)基因座的基因频率分布 ,得到群体遗传学数据。方法 用Chelex10 0快速法提取抗凝血的DNA ,经PCR扩增、变性聚丙烯酰胺凝胶电泳后 ,银染显带。统计各基因频率 ,计算杂合度、多态信息含量、个人识别概率及亲权否定概率。结果 得到 9个基因座各等位基因的频率 ,分布符合遗传平衡定律 ,具有高的杂合度和多态信息含量。个人识别概率 3组分别为 0 .9972、0 .9980和 0 .9996 ,3组累积个人识别概率为 0 .999999997。亲权否定概率 3组分别为 0 .85 6 5、0 .882 6和 0 .934 3,3组累积亲权否定概率为 0 .9989。结论  9个基因座具有较高的杂合度 ,是较理想的遗传标记 。 objective To investigate the allele frequencies of nine short tandem repeats(STR) divided into three groups(groupⅠ:CSF1PO, TPOX, TH01; groupⅡ:F13Ao1, FESFPS, vWA;groupⅢ:D16S539, D7S820, D13S317) in population of Han nationality in Henan province.Methods DNA extracted with Chelex100 from anti-coagulative blood sample was amplified with multiplex PCR. The PCR product was analyzed with the denatured polyacrylamide gel electrophoresis and silver-stain. After getting the allele frequencies, the heterozygosity (H), polymorphism information content(PIC), discrimination probability(DP) and exclusion power(PE) were calculated.Results All the nine allele frequencies were obtained and the frequencies agreed with Hardy-Weinberg equilibrium,and H and PIC of nine loci were high. The DP was 0.9972,0.9980 and 0.9996 respectively in three groups,the accumulated DP of three groups was 0.999999997, while the PE were 0.8565,0.8826 and 0.9343 respectively,and accumulated DP of three groups was 0.9989 Conclusion The nine STR loci prossesses high heterozygosity and are useful genetic marker,and can be used in personal identification and paternity test.
