目的 探讨 T- 2毒素对软骨细胞 IL- 1β与 IL- 6分泌的调节作用。方法 采用体外原代单层培养的人胎儿骺软骨细胞 ,T- 2毒素在有或无佛波醇酯 (PMA)刺激下与软骨细胞共同孵育。用酶联免疫吸附测定法(EL ISA)检测上清液中 IL- 1β与 IL- 6水平。结果 T- 2毒素为 8μg/L 时 ,不论有无 PMA刺激 ,IL- 1β分泌均显著增加 ;有 PMA刺激 ,IL- 6分泌显著增加。结论 T- 2毒素可以超诱导软骨细胞 IL- 1β与 IL-

Objective To investigate the effects of in vitro exposure to T-2 toxin on IL-1β and IL-6 secretion in human fetal chondrocytes.Methods The effects of exposure to t-2 toxin on Il-1β and IL-6 secretion were evaluated in primary monolayer culture of human fetal epiphyseal chondrocytes with or without phorbol 12-myristate 13-acetate (PMA) stimulation. The levels of supernatant IL-1β and IL-6 were analyzed by ELISA.Results Significant increases were seen in supernatant Il-1β and Il-6 obtained from 24 h PMA-stimulated chondrocyte cultures, and in supernatant Il-1β obtained from 48 h chondrocyte cultures without PMA stimulation containing T-2 concentration of 8 μg/L,compared with the corresponding controls.Conclusions The results demonstrate that T-2 toxin can superinduce Il-1β,Il-6 secretion in chondrocytes, All these data suggest that superinduction may be the key mechanism of chondrocyte injury by T-2 toxin.
