摘要
目的 研究氟对大鼠脑、肝、肾组织细胞凋亡和细胞周期的作用特点与氧化侵袭之间的关系 ,进一步探讨氟中毒发生机理。方法 在常规食、偏食条件下 ,经饮水投不同剂量氟 ,应用流式细胞仪方法检测脑、肝、肾组织细胞凋亡 ,细胞周期与活性氧产生状态。结果 常规食投氟 10 0~ 15 0 mg/ L、偏食投氟 5 0~ 10 0 mg/ L对脑、肝、肾组织活性氧产生无明显影响。氟对脑组织神经细胞周期的影响主要在 S期 ,可明显减少 S期神经细胞数量 ;对肝细胞周期的作用主要在 G1 期和 G2 期 ,偏食投氟各组 G1 期细胞数目普遍低于常食投氟组 ,G2 期细胞数目则高于常规投氟组。过量氟对肾组织的作用 ,主要为偏食投氟 10 0 mg/ L情况下 ,可明显诱导细胞凋亡的变化。结论 过量氟对细胞周期的影响 ,对不同组织的作用时相似有一定程度的差别 ,对脑神经细胞主要作用于 S期 ,肝细胞的影响主要是 G1 、G2 期 ;
Objective To investigate the effects of fluorosis on the cell cycle , cell apoptosis , and active oxygen of brain, liver, and kidneys in rats and discuss the mechanism of fluorosis.Methods In stock diets and unnutritive diets condition,drinking water was supplemented with different doses of fluoride. The cell cycle, cell apoptosis, and active oxygen in brain, liver, and kidneys of rats were inspected by flow cytometry.Results In stock diets 100~150 mg/L F groups,active oxygen in brain, liver,and kidneys of rats had no obvious change(P>0.05);the roles of fluorine in brain cell cycle appeared to be in S stage, the nervous cell counts in S stage of brain statistically decreased; compared to stock diet+F groups, the G 1 stage liver cell counts in unnutritive diets groups were obviously lowed and the G 2 stage of those were rised (P<0.05~0.01);the effects of fluorosis on kidneys mainly were to induce the apoptosis in unnutritive diets +100 mg/L F group (P<0.01). Conclusions The research results suggested the effects of fluorosis on those tissue appeared to be in S stage of brain, in G 1 and G 2 stage of liver, and in cell apoptosis of kidneys tissue.
