摘要
研究应用基因组原位杂交、染色体 C-分带和 RAPD技术 ,对八倍体小黑麦×普通小麦杂种 F2 经电离辐射处理后的高代材料 98- 60进行了检测。基因组原位杂交结果表明 ,该材料为小麦 -黑麦异代换系。进一步通过 C-分带分析表明 ,该品系为 5R代换系 ,并且还包含有5AS/ 6AS小麦种内的染色体易位。通过 RAPD分析 ,在该品系中找到了来源于八倍体小黑麦亲本“新麦 73”的黑麦染色体特异扩增产物 OPA- 0 1 350 和与两个亲本不同的特异重组产物OPF- 1 480 0 、OPF- 1 492 0 ,进一步验证了基因组原位杂交和
GISH,chromosome C banding and RAPD techniques were used in the detection of rye chromatin in high generation offspring (98 60) of radiation treated F 2 of crossing between octoploid triticale and common wheat.According to the result of GISH,these offspring are substitution lines,further analysis by C banding reveal the existence of 5R chromosome and a translocation of 5AS/6AS.These were also confirmed by RAPD that a amplified fragment,OPA 01 350 ,derives from octoploid triticale line “Xinmai 73” and two amplified recombination products,OPF 14 800 and OPF 14 920 ,are different from genomic DNA of two parents.
