摘要
以MS培养基上培养的银杏高产细胞系MH 3为材料 ,SH培养基为出发培养基 ,通过优化营养元素和激素配比 ,建立银杏致密细胞团悬浮培养体系 ,在培养过程中添加前体异戊二烯和香叶醇有助于提高银杏内酯产量。结果表明 :颗粒粒径大小影响银杏内酯的产生 ,选用 0 8SH培养基 ,添加浓度为 3mg/L的 2 ,4 D和 2mg/L的 6 BA ,致密颗粒的平均粒径为 3 36mm ,细胞中银杏内酯的含量为 5 5 7μg/g·dw。在培养过程中的 10d添加 10 0mg/L的香叶醇 ,细胞中银杏内酯的含量达到 6 76
The suspension culture system of compact cells group aggregates(CCGA)in Ginkgo biloba was established by using cell line MH 3 as experimental material,SH medium as basic medium,optimizing nutritional components and hormones combination.Adding precursors can profited ginkgolides formation during culture time.Results shows:the size of aggregates influence the ginkgolides content,when using 0.8 SH medium,adding 2,4 D(3 mg/L)and 6 BA(2 mg/L)to medium,the average size of aggregates was 2.71 mm,ginkgolides content in CCGA was 557 μg/g·dw.adding geraniol(100 mg/L)to medium on 10th day,ginkgolides content in CCGA was increased to 676 μg/g·dw.
