摘要
目的 探讨诱导新生大鼠免疫耐受的方法。方法 分别给新生SD大鼠胸腺或腹腔内注射 2 .5× 10 7个Wistar大鼠脾淋巴细胞 ,8~ 10周龄时行Wistar及第三品系大鼠供心移植。结果 胸腺或腹腔注射Wistar大鼠脾淋巴细胞者移植Wistar大鼠心脏后移植物的存活时间显著延长 ,胸腺注射者平均存活时间超过 60 .4d(P <0 .0 0 1) ,腹腔注射者平均存活时间为 15 .8d(P <0 .0 2 ) ,而第三品系大鼠的心脏移植后平均存活 9.2d(P >0 .0 5 )。结论 单一胸腺或腹腔内注射同种脾细胞可以诱导新生大鼠的特异性免疫耐受。
Objective To induce immunologic tolerance to cardiac grafts in neonatal SD rat. Methods Intrathymic (IT) or intraperitoneal (IP) injection of 2.5 ×10 7 Wistar spleen cells in neonates was followed by heterotopic cardiac transplantation using Wistar or the third party allograft at 8 to 10 weeks of age. Results The survival time of Wistar cardiac grafts was prolonged with both intrathymic or intraperitoneal inoculation of allogeneic cells (IT: > 60.4 ?days, Р < 0.001 ; IP: 15.8 ?days, P < 0.02 ). Third party allografts were rejected in the usual time frame ( 9.2 ?days, P > 0.05 ). Conclusions Intrathymic or intraperitoneal inoculation of allogeneic cells in a neonatal recipient with an immature system can produce donor specific tolerance to a subsequent graft.
