不同年龄小鼠染色体DNA与4.5sRNA逆转座子同源序列杂交的变化 被引量:2

摘要 目的 研究不同年龄小鼠脑基因组 DNA中 4.5s RNA逆转座子同源序列含量的变化。方法 用生物素化补骨脂素标记的 p SVLuc-4 .5s质粒 DNA(携带 4.5s RNA逆转座子序列 )作为探针 ,提取不同年龄的小鼠脑组织染色体 DNA与探针进行斑点杂交。结果 随着小鼠年龄的增加 ,同源序列的拷贝数减少。结论  4.5s RNA逆转座子以及小鼠基因组中的大量短分散重复序列在个体发育过程中是处于动态变化 ,与衰老过程密切相关。 Objective To determine th e content difference of the homologous sequence of 4.5s retroposon RNA in mice ge nomes of different ages.Methods Biotinated psoralen(B-P)was used to label the pl asmid pSVLuc-4.5s(carrying retroposon 4.5s DNA)as the probe.Dot hybridization between the probe and the chromosomal DNA from mice of different ages was conduc ted in order.Results It was shown that with the increase of age,copy number of homologous sequence decreases.Conclusions 4.5s retroposon RNA changes with growth and shows close relation with a geing.
