摘要
目的 :新建一株人肝癌细胞系 ,为肝癌研究提供新的实验模型。方法 :用组织块法培养 ,并按细胞系建立标准检测细胞的一系列生物学特性。结果 :细胞维持培养 15个月 ,传代 10 0代 ,细胞形态及超微结构具有恶性细胞的特征 ,45代细胞的倍增时间为 2 1 3h ,细胞能在软琼脂上生长 ,形成集落 ,染色体众数60～ 67条 ,主干系为亚三倍体。 1q(i) 和t(6,11)为其标记染色体 ,细胞对刀豆球蛋白有凝集反应 ,5× 10 6 细胞接种于裸鼠皮下 ,肿瘤呈进行性生长。结论 :该细胞系符合建系标准 ,是一株新建的肝癌细胞系。
Objective:To establish a new hepatocellular carcinoma (HCC) cell line and provide a new experiment model for the research of HCC.Methods:The cell line was cultured in vitro and a series of characteristics of the cells was examined according to the rules of establishing a cell line in vitro after the primary cells were successively passaged.Results:The cell line designated as HCC 9903 had been so far maintained for 15 months through 100 passages,morphological and ultrastructure observations demonstrated that the cells corresponded well with the malignant epithelioid cell morphology.The doubling time was 21.3 hours at passage 45.The cells grew in soft agar,chromosome analysis revealed that it had a wide variation mode of around 60～67,aberrant chromosomes lq (i) and t(6,11) were its chromosome markers,celm agglutination was observed when treated with Con A,tumor was taken when 5×10 6 cells of HCC 9903 suspension were transplanted into the nude mice.Conclusion:According to the rules of establishing a cell line in vitro,HCC 9903 is a new cell line of human hepatocellular carcinoma.
