肺癌Th1/Th2反应状态及黄芪的调节作用 被引量:17

摘要 目的 :探讨肺癌细胞株以及荷瘤机体外周血单个核细胞 (PBMC)Th1/Th2免疫反应状态 ,观察中药黄芪 (AG)对两者Th1/Th2状态的作用。方法 :① 3种人肺癌细胞株 (鳞、腺、小细胞癌 )各分为 5组 ,其中 1组即刻冻存待测 ,其余 4组分别为不加药对照组、加 5 %AG、10 %AG、2 0 %AG组 ,培养 48h ,收集细胞 ;②采集并分离肺癌患者和健康志愿者的PBMCs,各分为 4组 :其中 1组即刻冻存待测 ,其余 3组分别为不加药对照组、加 10 %AG、2 0 %AG组 ,培养 48h ,收集细胞。以IL 2 ,IFN γ代表Th1型细胞因子 ,IL 4,IL 6,IL 10代表Th2型细胞因子 ,用RT PCR方法检测各组肺癌细胞株和肺癌患者PBMC中Th1/Th2型细胞因子m RNA的表达。结果 :① 3种肺癌细胞株的Th1型因子IFN γ和IL 2均无表达 ,Th2型因子IL 4、IL 6、IL 10则均表达 ;经与黄芪培养后 ,小细胞肺癌株在 3种不同浓度AG存在时均可出现IFN γ表达 ,在5 %AG时可同时出现IL 2的表达 ,IL 4、IL 6的表达被抑制 ;鳞癌和腺癌细胞株经黄芪培养后 ,其Th1/Th2型细胞因子的m RNA表达与培养前差异无显著性 (P >0 0 5 ) ;②肺癌患者PBMC中 ,IL 4、IL 6和IL 10表达阳性率显著高于正常对照 (P均 <0 0 5 ) ,而IL 2无 1例表达 (0 /2 3 ) ,IFN γ仅 1例表达 ;经与黄芪培养后(两种浓度 ) ,? Objective:To investigate the immunological reaction state of Th1/Th2 cytokines expression in human lung cancer cell lines and the peripheral blood mononuclear cells(PBMCs) from patients with lung cancer,and evaluate the effect of astragalus(AG) on them.Methods:The cells from three kinds of human lung cancer cell lines were divided into 5 groups:one group as pre cultured group,and the other cells were cultured with 5% AG,10% AG,20% AG and without AG(control group),respectively.The PBMCs from patients were divided into 4 groups:one group as pre cultured group,and the other cells were cultured with 10% AG,20% AG and without AG (control group),respectively.Three kinds of human lung cancer cell lines and PBMC from 23 lung cancer patients as well as 19 healthy controls before and after the culture with AG were collected respectively.The Th1/Th2 cytokines expression were detected by RT PCR considering IFN γ and IL 2 as Th1 cytokines and IL 4,IL 6,IL 10 as Th2 ones.Results:None of the human lung cancer cell lines expressed IL 2 and IFN γ,whereas IL 4,IL 6,IL 10 were expressed in pre cultured and control groups.After the culture with AG,the expression of IFN γ was induced,while that of IL 4,IL 6 was inhibited,especially in SCL.The m RNA expression rates of IL 4,IL 6,IL 10 in PBMCs from lung cancer patients were obviously higher than those from the healthy controls ( P<0.05) ?IL 2 expression was not detected in all patients(0/23),and the positive rate of IFN γ expression was 1/23;After being cultured with AG,the expressions of Th1/Th2 cytokines were changed obviously ,which was refleted by a significant enhancement in IL 2 expression paralleled by a substantial decrease in IL 4,IL 6 and IL 10.Conclusion:The expression of Th1 cytokines was inhibited with a concomitant predominant state of Th2 cytokines in both human lung cancer cell lines and PBMCs from lung cancer patients.AG can convert the imbalant state of Th1/Th2 cytokines and make it incline to the normal state.
