摘要
目的 探讨环孢素A(CsA)慢性肾毒性的发生机理。方法 给予进正常饮食或低盐饮食的大鼠灌服CsA 3 0mg·kg-1·d-1,采用逆转录聚合酶链反应 (RT PCR)研究CsA对大鼠肾内转化生长因子 β1(TGF β1)及肾素mRNA表达的影响。 结果 给予CsA的大鼠均有肾内TGF β1mRNA、肾素mRNA表达的增加 ;给予低盐饮食的大鼠这些改变更为明显。结论 CsA的慢性肾毒性与肾内TGF β1mRNA、肾素mRNA表达的增加有关。
Objective To explore the mechanisms of cyclosporin induced chronic nephrotoxicity. Methods Rats were on low salt diet or normal diet and cyclosporine A (CsA) was administered once a day by gastric gavage at doses of 30?mg·kg -1 ·d -1 for 28?days. The expression of mRNA for intrarenal transforming growth factor β1 (TGF β1) and renin was detected by reverse transcription polymerase chain reaction (RT PCR). Results Increased expression of TGF β1 and renin mRNA was found in the rats receiving CsA both on low salt diet and normal diet. The increase was more obvious in the rats on low salt diet. Conclusion Cyclosporine induced chronic nephropathy may be related to the up regulation of TGF β1 and renin mRNA.
