摘要
从鸡胚发育为 X期的鸡蛋中取胚盘细胞 ,经原代培养后用带增强型绿色荧光蛋白( EGFP)作为标记基因的真核表达质粒 p L EGFP-C1转染鸡胚细胞观察 EGFP基因的表达情况 ,结果在不同条件下的活细胞均见有 EGFP基因的稳定表达。这提示通过 EGFP基因转染鸡胚盘细胞可连续且直接观察活细胞中的表达 。
To search the appropriate experimental condition for using green fluorescent protein (GFP) as a reporter in transgenic chicken, Freshly laid unincubated eggs from White Leghorn Chicken were used. Only embryos at stage X were used to separate the blastodermal cells. After incubated over night ,the blastodermal cells were transfected with pLEGFP C1 by lipofectame. Fluorescence could be detected in intact viable cells under different sets of conditions, and the expressed fluorescence was stable. The transfection rate of pLEGFP C1 expressed was different in three cells lines examined. The GFP can be visualized continuously and directly for gene expression in living cells.
