摘要
对碳源、氮源种类和用量对转rhTNF -α基因鱼腥藻 712 0 (Anabaenasp .PCC712 0 )培养的影响进行了研究 ,发现最适碳源为蔗糖 ,最适氮源为NaNO3 ,最佳用量分别为 9g/L和 2 .2 5g/L ,此时生物量远高于自养方式 ,达 2 .5 2 g/L ,比相同条件下在BG - 11培养基培养高 71.66%,TNF-α表达量为 16%~ 2 2 %,生物活性为 10 5U /mg。
The effects of carbon and nitrogen sources on the culture of Anabaena sp. PCC7120 harboring pDC-TNF were studied. The results showed that sucrose and NaNO 3 were the optimum carbon and nitrogen sources. When 9 g/L of sucrose and 2.25 g/L of NaNO 3 were used, the biomass was 2.52 g/L which was 71.66% greater than culturing in BG-11 under the same condition, and the expression and bioactivity of TNF were 16%~22% and 10 5 U/mg, respectively.
