摘要
目的 探讨观察重组人内抑素对人血管内皮细胞增殖影响的方法。方法 采用MTT法观察不同浓度重组人胎儿内抑素是否对碱性成纤维细胞生长因子刺激的人血管内皮细胞系ECV增殖产生抑制作用。结果 MTT法分析显示细胞活力与光密度改变之间存在线性关系 ,重组人内抑素可抑制胎儿血管内皮细胞ECV的增殖 ,半数抑制浓度约为 3 5 0mg·L-1。结论 MTT法检测重组人内抑素对人血管内皮细胞增殖的抑制作用准确可靠、操作简便。
Objective To investigate method to assay effect of recombinant human enostatin (rhEndo)on proliferation of human vascular endothelial cells. Methods MTT method was used to observe roles of recombinant human endostatin of different concentrations on the proliferation of fetus endothelial cell line ECV. Results MTT assay showed that cell viability was correlated positively with optical density and rhEndo could inhibit the proliferation of endothelial cell ECV. It demonstrated the most obvious inhibitory effect at the concentration of 20 mg·L -1 , and the ED 50 is about 3 50 mg·L -1 . Conclusion MTT method can be used to analyze the inhibitory roles of rhEndo expressed by E. Coli BL 21(DE 3)SI on proliferation of basic fibroblast growth factor(bFGF) stimulated human vascular endothelial cells, which is reliable, simple and convenient.
