摘要
观察腹腔注射细菌内毒素 (LPS)后 ,大鼠延髓内脏带 (MVZ)星形胶质细胞胶质原纤维酸性蛋白 (GFAP)及神经元FOS表达水平随时间变化的规律及其相互关系。大鼠经LPS腹腔注射后 1,3,6,12h分别行固定取材制片。每个时间点的切片分为 3组 ,分别进行抗FOS、抗GFAP免疫组织化学染色及抗FOS/GFAP/酪氨酸羟化酶 (TH)三重免疫组织化学染色。结果表明 :①FOS反应在LPS注射后 3h达到高峰 ,阳性产物主要分布于MVZ内。②GFAP反应在注射后 1h即达到高峰 ,表现为胶质细胞肥大、数量增多。其分布与FOS基本相同。③三重染色观察到GFAP与FOS的多种聚集方式 (FOS/GFAP/TH ,FOS/GFAP ,GFAP/TH) ,FOS阳性神经元周围GFAP免疫反应产物更密集。提示星形胶质细胞对LPS起反应 。
To investigate the effect of lip op olysaccharide (LPS) on the time dependent expressions and relationship of glial fibrillary acidic protein (GFAP) and FOS in the medullary visceral zone (MVZ) o f the rat, anti GFAP and anti FOS immunohistochemical single and anti FOS/G FAP/tyrosine hydroxylase (TH) triple labeling methods were used to observe the medullary oblongata of the rats at 1,3,6,12 h after intraperitoneal (i.p.) LPS i njection. The results indicate that ①Fos positive neurons were mostly found in MVZ and reached the peak value at 3 h. ②An increase in the number and size of G FAP positive astrocytes was found after i.p. LPS. It reached the peak value at 1 h which is earlier than FOS. At the same time, the distribution of GFAP was sim ilar to FOS. ③With triple labeling method, three kinds of the complexes were f ound including FOS/GFAP/TH, FOS/GFAP and GFAP/TH. It is concluded that astrocyt es react to LPS and the peak value of GFAP is prior to that of FOS, suggesting that astrocytes are another important group with a variety of functions in the central nervous system.
