端粒酶活性在脑胶质瘤中的检测 被引量:1

摘要 目的 :通过检测脑胶质瘤中端粒酶的表达活性及肿瘤增殖标记抗原 Ki-67的表达 ,初步探讨端粒酶活性水平与脑胶质瘤的相关性及临床意义。 方法 :采集 4 0例脑胶质瘤手术切除标本、4例正常脑组织 ,通过半定量端粒重复序列扩增 ( TRAP)银染法检测端粒酶活性水平 ;采用免疫组化法检测胶质瘤标本的增殖活性标记抗原 Ki-67。 结果 :4 0例胶质瘤标本中有 3 3例 ( 82 .5% )检出端粒酶活性 ,而正常脑组织中无端粒酶活性表达 ;不同级别胶质瘤之间端粒酶活性水平有明显差异。端粒酶活性高低与 Ki-67的表达呈显著正相关 ( r=0 .74 5,P<0 .0 5)。 结论 :提示端粒酶活性可作为脑胶质瘤的恶性程度标记之一。 Objective: : To study the activity of telomerase in patien ts with glioma and its relationship with glioma. Methods: Forty fr eshly resected glioma specimens and 4 normal brain tissue were studied. Telomera se activity was detected by semi-quantitative telomeric repeat amplification pr otocol (TRAP) through silver-staining. The proliferative activity of gliomas wa s also investigated by Ki-67 immuno-labelling. Results: Telomerase activity was detected in 33 of 40(82.5%) glioma specimens and was not detected in normal brain tissue, telomerase expression was positively associated with gra de of tumor and Ki-67 immuno-labelling( r =0.745, P <0.05). Conclus ion: Telomerase activity can be a special marker of gliomas and an index f or malignant potential.
