摘要 本研究利用DNA重组技术构建了来源于质粒 pJBJ10 6、pBluescriptSK(+ )和pUCD80 0 的新质粒 pXT3sacB ,利用该质粒通过同源重组切除了野生型发根农杆菌K599中Ri质粒的T DNA。解毒后的K599获得了氨苄 /羧苄青霉素抗性和 10 %蔗糖抑制生长的选择标记。解毒后的K599菌株可能对农杆菌转基因技术是有益的。 In this study, the new plasmid pXT 3sacB was constructed from pJBJ 106 ,pBluescript SK(+) and pUCD 800 . The wild type T DNA of Agrobacterium rhyzogenes K 599 which induces hair root was deleted with pXT 3sacB by homologous recombinant. A disarming K 599 contains selective markers of resistance to ampicillin/carbencillin and the growth inhibitation by 10% sucrose. It could be a novel useful strain for gene transfer via Agrobacterium-mediated method.
