摘要
目的观察 7-氯苄基四氢巴马汀 (7-chlor -BTHP)对正常及肥厚心脏大鼠血管环的作用。方法用去甲肾上腺素 (NA)和氯化钾 (KCl)引起血管收缩 ,然后观察 7-chlor -BTHP对正常及肥厚心脏大鼠血管环的作用。结果在肥厚及正常心脏大鼠的离体血管环上 ,7-chlor-BTHP及奎尼丁 (Qui)对NA所致的收缩均有抑制作用。 7-chlor -BTHP的IC50 分别为 76及 3 9μmol·L- 1,而Qui则分别为4 .6及 8μmol·L- 1。Qui对氯化钾所致血管环的收缩有明显的抑制作用 ,其PD2 ’分别为 4 .3 (肥厚 )及 4 .0 (正常 ) ,而 7-chlor-BTHP则对其无影响。 结论 7-chlor-BTHP有拮抗α -受体作用 。
Objective To observe the effects of 7-chlor-benzyltetrahydropalmatine(7-chlor-BTHP) on isolated aortic ring from normal and hypertrophic heart of rats.Methods The isolated aortic rings from normal and hypertrophic heart of rats were prepared and excited by NA and KCl,then to observe the infulence of 7-chlor-BTHP on these aortic rings.Results Both 7-chlor-BTHP and quinidine were showed the blocking effect on α adrenoreceptor in aortic rings from two kinds of heart and could reduce contraction induced by norepinephrine in concentration dependent manner,the IC 50 of 7-chlor-BTHP (76μmol·L -1 for hypertrophic heart and 39μmol·L -1 for normal) were much higher than those of quinidine (4.6 μmol·L -1 for hypertrophic heart and 8μmol·L -1 for normal).The concentration response to KCl was non competitively inhibited by quinidine in aortic rings from normal and hypertrophic hearts,the PD 2′valves were 4.0 and 4.3 respectively,but 7-chlor-BTHP was unable to shife the KCl concentration respone curves,indicating that it had no influence on potential dependent calcium channel.Conclusion The results indicate that 7-chlor-BTHP can block α adrenoreceptor,but can not block calcium channel.
