摘要
将经消化后的新西兰幼兔软骨细胞接种在含有碱性成纤维细胞生长因子 (bFGF)或缺乏bFGF的培养液中进行培养 ,观察bFGF对软骨细胞的增生作用及其最佳质量浓度 ,并测取软骨细胞对3 H 胸腺嘧啶的摄取量。发现bFGF可促进软骨细胞数目增多 ,促进软骨细胞增生的最佳质量浓度为 1 0~ 1 5 μg/L ;培养基中含有bFGF和缺乏bFGF生长的软骨细胞对3 H 胸腺嘧啶摄取量有明显差异 (P <0 .0 5 )。提示 :bFGF可促进兔关节软骨细胞增生 ,其促细胞增生作用在一定范围内同bFGF质量浓度成正相关。
The purpose of this study is to investigate the effect of bFGF on rabbit articular chondrocyte culture in vitro, so that the study could help search a clue of treatment for repair of cartilagious injury. Chondrocytes were isolated from articular cartilage of 1~3 d old New Zealand rabbit. Condrocytes were seeded at different densities of in 10 mL 1640 medium with different concentration of bFGF, to observe the proliferation effects of bFGF on chondrocytes and the best concentration of bFGF for stimulatory proliferation and to measure the amount of incorporation of 3 H thymidine. The amount of chondrocyte increased with increase of concentration of bFGF and the best concentration of bFGF was 10~15 μg/L for stimulatory proliferation. The amount of incorporation of 3 H thymidine was greater in chondrocyte culture grown with bFGF than that tacking bFGF( P <0.05). bFGF exerts a strong effect on proliferation of cultured chondrocytes and the best concentration of bFGF was 10~15 μg/L.
